pDNA and mRNA bioprocessing: theory and practice

About course
In this 4-day, hands-on course, you will get a comprehensive understanding of the mRNA workflow, starting from downstream purification of plasmid DNA (pDNA).
You will get practical experience on how to purify pDNA, starting from cell lysis to the polishing chromatography step. You will then learn how to perform an IVT reaction and investigate the impact that certain parameters have on the mRNA yield. Understand mRNA purification using Fibro™ OligodT and the effect of parameters such as salt concentration and residence time.
Participants will get to know the plasmid DNA and mRNA end-to-end workflow through classroom lectures.
At the end of this course, you will be able to:
- Understand the different unit operations in plasmid DNA and mRNA manufacturing processes
- Gain practical experience in pDNA purification, mRNA synthesis and mRNA purification
- Understand process hygiene, IVT, LNP, and scale-up considerations
- Identify key analytical methods required to assess mRNA quality
